(一)酸度计原理
用酸度计进行电位测量是测量pH最精密的方法。pH计由三个部件构成：
(1)一个参比电极；
(2)一个玻璃电极，其电位取决于周围溶液的pH；
(3)一个电流计，该电流计能在电阻极大的电路中测量出微小的电位差。由于采用最新的电极设计和固体电路技术，现在最好的pH可分辨出0.005pH单位。参比电极的基本功能是维持一个恒定的电位，作为测量各种偏离电位的对照。银-氧化银电极是目前pH中最常用的参比电极。玻璃电极的功能是建立一个对所测量溶液的氢离子活度发生变化作出反应的电位差。把对pH敏感的电极和参比电极放在同一溶液中，就组成一个原电池，该电池的电位是玻璃电极和参比电极电位的代数和。E电池＝E参比+E玻璃，如果温度恒定，这个电池的电位随待测溶液的pH变化而变化，而测量pH计中的电池产生的电位是困难的，因其电动势非常小，且电路的阻抗又非常大(1－100MΩ)；因此，必须把信号放大，使其足以推动标准毫伏表或毫安表。电流计的功能就是将原电池的电位放大若干倍，放大了的信号通过电表显示出，电表指针偏转的程度表示其推动的信号的强度，为了使用上的需要，pH电流表的表盘刻有相应的pH数值；而数字式pH计则直接以数字显出pH值。
(二)酸度计调试　
实验室常用的pH计有老式的国产雷磁25型酸度计(最小分度0.1单位)和pHS-2型酸度计(最小分度0.02单位)，这类酸度计的pH值是以电表指针显示。新式数字式pH计有国产的科立龙公司的KL系列，其设定温度和pH值都在屏幕上以数字的形式显示。无论哪种pH计在使用前均需用标准缓冲液进行二重点校对。
首先阅读仪器使用说明书，接通电源，安装电极。在小烧杯中加入pH值为7.0的标准缓冲液，将电极浸入，轻轻摇动烧杯，使电极所接触的溶液均匀。按不同的pH计所附的说明书读取溶液的pH值，校对pH计，使其读数与标准缓冲液(pH7.0)的实际值相同并稳定；然后再将电极从溶液中取出并用蒸馏水充分淋洗，将小烧杯中换入pH4.01或0.01的标准缓冲液，把电极浸入，重复上述步骤使其读数稳定。这样就完成了二重点校正；校正完毕，用蒸馏水冲洗电极和烧杯。校正后切勿再旋转定位调节器，否则必须重新校正。
(三)酸度计使用
所测溶液的温度应与标准缓冲液的温度相同。因此，使用前必须调节温度调节器或斜率调节旋钮。先进的pH计在线路中安插有温度补偿系统，仪器经初次较正后，能自动调整温度变化。?测量时，先用蒸馏水冲洗两电极，用滤纸轻轻吸干电极上残余的溶液，或用待测液洗电极。然后，将电极浸入盛有待测溶液的烧杯中，轻轻摇动烧杯，使溶液均匀，按下读数开关，指针所指的数值即为待测溶液的pH值，重复几次，直到数值不变(数字式pH计在约10s内数值变化少于0.01pH值时)，表明已达到稳定读数。测量完毕，关闭电源，冲洗电极，玻璃电极要浸泡在蒸馏水中。
(四)酸度计保养及注意事项
玻璃电极在初次使用前，必须在蒸馏水中浸泡一昼夜以上，平时也应浸泡在蒸馏水中以备随时使用。玻璃电极不要与强吸水溶剂接触太久，在强碱溶液中使用应尽快操作，用毕立即用水洗净，玻璃电极球泡膜很薄，不能与玻璃杯及硬物相碰；玻璃膜沾上油污时，应先用酒精，再用四氯化碳或乙醚，最后用酒精浸泡，再用蒸馏水洗净。如测定含蛋白质的溶液的pH时，电极表面被蛋白质污染，导致读数不可靠，也不稳定，出现误差，这时可将电极浸泡在稀HCl（0.1mol/L）中4－6分钟来矫正。电极清洗后只能用滤纸轻轻吸干，切勿用织物擦抹，这会使电极产生静电荷而导致读数错误。甘汞电极在使用时，注意电极内要充满氯化钾溶液，应无气泡，防止断路。应有少许氯化钾结晶存在，以使溶液保持饱和状态，使用时拨去电极上顶端的橡皮塞，从毛细管中流出少量的氯化钾溶液，使测定结果可靠。
另外，pH测定的准确性取决于标准缓冲液的准确性。酸度计用的标准缓冲液，要求有较大的稳定性，较小的温度依赖性。
